Instrumentation for UHPLC

Abstract This chapter explains the instrumentation needed to take full advantage of smaller particle packed columns, and what is needed to maintain a well functioning system under pressures above 6,000 psi (~400 bar). We also provide guidelines for proper instrumentation and parameter selection to run the best possible separations and to use the optimum columns of a specific dimension and particle size. We will then touch briefly on the effect of unwanted extra-column band spreading on column efficiency and how to minimize it.
